Extruder & Spheroniser for Pharmaceutical Pelletisation
Pelletisation is widely used in pharmaceutical manufacturing to produce uniform, spherical multi-particulates for modified release, coating and capsule-filling applications.
The Extrusion–Spheronisation process converts a wet granulated mass into controlled, spherical pellets with good flow, narrow particle-size distribution and suitable mechanical strength.
Extrusion
In the extrusion stage, the wet mass is passed through a defined screen or die to form uniform cylindrical extrudates.
The extrusion process helps control:
- Extrudate diameter
- Density
- Moisture distribution
- Uniformity
- Feed consistency
The characteristics of the wet mass and extrusion parameters directly influence the quality of the final pellets.
Spheronisation
The extrudates are transferred to the Spheroniser, where they are broken and rounded into spherical pellets through controlled rotational movement.
Spheronisation helps achieve:
- High sphericity
- Uniform pellet size
- Smooth surface
- Good flowability
- Controlled density
- Reduced fines
These characteristics are important for consistent coating and downstream processing.
Why Pellet Uniformity Matters
Uniform pellets provide a reliable base for subsequent pharmaceutical processing.
They can support:
- Consistent drug distribution
- Uniform functional coating
- Better capsule filling
- Improved flow during handling
- Controlled or modified drug release
Pellet quality depends on both formulation characteristics and process parameters.
Important Process Considerations
Successful extrusion and spheronisation depend on factors such as:
- Wet mass consistency
- Binder concentration
- Moisture level
- Extrusion screen size
- Extrusion speed
- Spheronisation speed
- Residence time
- Batch load
- Desired pellet size
Optimisation of these parameters helps achieve the required pellet size, shape and mechanical properties.
Integration with Downstream Processing
After spheronisation, pellets can be dried and further processed depending on the formulation requirement.
They may undergo:
- Drying
- Sieving
- Functional coating
- Drug layering
- Modified-release coating
This makes extrusion–spheronisation an important part of integrated pharma pellet manufacturing lines.
